I am working on extracting proteins from E. coli cells. However, the native proteins are present in very small quantities, making extraction challenging and increased risk of yield loss. To minimize this loss, I want to standardize the minimum expected protein yield, for more consistent and reliable results.
Have you thought about overexpressing your protein using a plasmid and then purifying it? I guess this would solve your issue
Why not scale up the amount of E. coli you are growing to compensate for low yields and purification losses. Look up publications on how E. coli DNA polymerases were purified from E. coli.
A better method to purify proteins in very small quantities is the affinity chromatographic technique. The interaction between the target molecule and the ligand must be reversible and safe to preserve the native protein properties.
