I developed some small peptide biomimetic compounds with different metallic centres, but I am having problems to purify it. The biggest problem is that it is only soluble and stable in DMSO. The compounds have about 2kDa. I was thinking in using columns, but at the same time all this work have to be done inside of a glovebox because of the high oxygen sensitivity of those compounds. Do any of you have any idea of how I can purify this?
Hi Sergio, I assume you want to purify the complex from a mixture of your (HPLC-purified?) peptides and the metal precursors. Given the oxygen sensitivity I don't see any option with standard techniques, but since the compounds are only soluble in DMSO I'd try precipitation/crystallization as the obvious procedure.
Hi thank you for you answer by now. There is another big information that I forgot to tell, they are completely unstable in water (even at really small amounts).
So, you don't have many options. is MeOH OK?. How polar are those compounds?, would it be possible to do a small flash column?. There are also disposable reverse-phase cartridges that you could probably run with a syringe, or perhaps pack a small plastic column with a reverse-phase phase and run the column by gravity... but I still think that crystallisation is the way to go here. At least to remove the soluble byproducts and get the crude cleaned up a bit. good luck
