due to colonies/agglomerates Streptomyces spp. forms in broth, I cannot use standard simple techniques such as OD600 or viable count to check isolates' development over time or survivability in stress conditions. Or at least I can but I don't think the data I obtain is realistic, especially not the OD.
Does anyone know how to solve this? or if there is a standard way to do that with Streptomyces?

You can use a DNA-based quantitation method like « quantitative real-time polymerase chain reactions (PCR) using primers designed and based on the DNA sequence of your Streptomyces species of interest ».
