I have tried fixing in PFA 4% for an hour followed by 2 PBS washes, it has worked but not all the times and I end up losing valuable samples. I have seen comments that indicate to do what I´ve been doing and add an acetone step 10 mins at -20°C. I would appreciate any other recommendations.

Hola Keren. Para tu investigación creo que puedes utilizar el protocolo que yo usaba para trabajar con tejido nervioso fijado en Formalina 10% con buffer de Sörensen pH 7, modificando algunos puntos.
1. Recoger los cortes del baño de flotación a 45°C (H2O-d) en portaobjetos cargados electrostáticamente (δ+)
2. Secar en estufa a 60°C x 1 hora mínimo
3. Xileno > EtOH 100 > Piroxilina (Colodión) 1% en EtOH 100+Eter etílico (1+1) > secar al aire > hidratar en H2O-d > Hematoxilina de Gill3 > H2O-d > virar hasta azul en NaHCO3 al 1% o Solución de Scott > H2O-d > Eosina 1% + HOAc (g) 0,05 ml (1 gota) > H2O-d > secado suave al aire > EtOH 100° > EtOH 100+Eter etílico (1+1) > Piroxilina (Colodión) 1% en EtOH 100+Eter etílico (1+1) > secar al aire > Xileno > montar cubreobjetos con Beta-pineno ± Xileno
Observaciones
a. La estructura de emparedado que se forma entre la carga electrostática del portaobjetos y la película de Piroxilina (Colodión) 1% en EtOH 100+Eter etílico (1+1) atrapa el corte impidiendo que se desprenda del medio de soporte.

b. El envase que contiene la mezcla de EtOH+Eter etílico (1+1) y la solución de Piroxilina al 1% disuelta en esta mezcla deben mantenerse a -20°C para evitar la formación de peróxidos y prevenir la evaporación. Los envases de polietileno (HDPE) de alta densidad (Nalgene) resultan muy apropiados. En el mercado están disponibles con sello hermético.
c. Los cortes pueden desprenderse durante el lavado en agua por el efecto disruptivo (formación de remolinos). Para evitarlo basta con regular el chorro de agua. La distancia a la que se encuentre la llave del tubo con respecto a la pileta de lavado es muy importante ya que causa además del efecto disruptivo, ondas de choque con desplazamiento horizontal.

d. Para trabajar con tejidos enclastrados en OCT debes modificar el protocolo a partir del punto 3, eliminando todo el proceso para cortes en parafina. Ingresar a partir de EtOH 100, luego de secar los cortes al aire.

It is difficult to answer properly without knowing details of the samples: a) size of the aortic sample; b) method of embedding in OCT; c) thickness of the sections; d) slides used; d) vertical or horizontal position of the slides during staining. It should be a question of adhesion if the section is directly collected on the slide from the plate, temperature of the chunk/slide may also be a factor.

A) Size: 10 um, b) I have samples that I embedded with the OCT on liquid nitrogen and others that were frozen to -20°, both storaged in -80°C. d) Corning slides with Poly-L-lisine. e) Vertical position while staining, horizontal in the ethanol & xylene steps. I collect the sample in a microtome at -22°C and the slides are at room temperature.

Hola Karen. Te adjunto ficha de seguridad y la descripción del producto en catálogo de dos proveedores. Es importante hacer referencia al número CAS 9004-70-0 ya que el término Piroxilina refiere el nombre de un componente de laca para madera y Colodíon, un excipiente de un producto farmacéutico antiverrucoso. I can identify two possible problems: a) 10 𝜇m is a thick section and may detach from the slide, 5 𝜇m is the usual thickness for histological examination; b) I never used liquid nitrogen because can alter the tissue properties, affecting subsequent cutting. I embed with the following steps: cold metal chunk, layer of OCT up to gel consistency, tissue section, cover with OCT, metal weight on top without pressing, freezing fluorocarbon spray on the metal weight. You should have a flat surface with tissue well integrated in the OCT. If cutting still difficult, gently adjust temperature with gloved thumb -22 might be a little too cold for certain tissues (I do not have experience with arteries) and you can try -18.
