I have a few questions about an optimal silica nanoparticles suspension, many authors use ethanol to suspend them before dilution for a cell stimulation and some other authors use Milli-Q water
why do nanoparticles (specially silica nanoparticles) are prepared in stock solutions using ethanol???
It is improper to suspend them directly in culture medium?
And, It's strictly necessary to use glass recipients for the stock solution preparation or it can be prepare in eppendorf tubes?
all depends on purpose and strategy. Silica is not used as stock solution (e.g. dissolved as silicate) but as stock dispersion preserving the particles. There are several silica products as dispersion in water, e.g. Ludox, Köstrosol, which can be directly dispersed in culture medium. Be aware that likely interaction with biomaterial will occur, which may or may not lead to flocculation.
I don't think there is an issue suspending them directly in culture medium. Eppendorfs should also be fine - if the volume is sufficient.
