How can I measure the sodium hydroxide content in a freeze dried sample? The sample was extracted from the raw material by using sodium hydroxide solution. Once the sample was extracted (soluble in NaOH solution), the solution was freeze dried. I worry that the sodium hydroxide was freeze dried as well. In my case, I can not apply acid titration since my sample also consists of an OH group.

If the OH in your sample is a bound hydroxyl group, and the only significant source of free OH- ions would be the NaOH, you could titrate with AgNO3 and weigh any insoluble AgOH that precipitates out.

If the OH is only source of your sample you can make quantitative analysis of OH by using FT-IR with ATR unite.
