1) you need to identify the longest ORF using an ORF finder/predictor software online
2) Convert that ORF sequence of mRNA to DNA using http://molbiol-tools.ca/Convert.htm (remember to make sure that if you are converting sequence into DNA for expression into E.coli, you MUST use the E.coli codon bias to translate mRNA to DNA).
3) Design appropriate primer with restriction sites/epitope tags in frame either N-terminally or C-terminally, to clone into bacterial expression vector/plasmid (eg. pET28 and others).
