I am planning to extract DNA from a plant mixture, I need to shake samples in Erlenmeyer flasks. I want to make sure they are free of any external pollution especially of bacterial DNA which will be separated from plant DNA in future and it is important.
This question also exists for falcon tubes. I want to know if they are newly bought, are they free of DNA or is there a procedure to make sure there is nothing external?
Wash the labware with 7M NaOH and rinse thoroughly with sterilize or autoclave water. You can also dry autoclave the labware before use. The plastic ware from a good company is generally NA free, but if you are not sure, you can do similar treatment or UV sterilization. Mostly, the contamination occurs due to the user than the unused plastic ware.
use DNA off trade mark buffer or the other DNase that companies sell for this purpose.
Really? Use DNase and the residue will digest DNA of interest, expensive on top of that. Are you sure you know what you are suggesting?
