How can I make a 50pmol/ul EcoRI adaptor?

Hello.

I am working on the AFLP analysis of C. acutatum fungi.

I need to make EcoRI adaptor ( 50pmol/ul ).

I have primer 1 and primer 2 that are somewhat complementary to each other to

generate EcoR1 adaptor.

Primer 1 is 100pmol/ul.

primer 2 is also 100pmol/ul.

Does it become 50pmol/ul EcoRI adaptor,

If I mix 50 ul of primer 1 and 50 ul of primer 2 ???

In theory, yes. The total volume would be 100ul giving a dilution factor of 0.5 for each. However, you may need to use a ligation buffer as well which would need to be taken into account for total volume.
