Hello,
I would like to try and locate a locus in the genome of a plant that confers a specific phenotype so that I can genotype plants as having either A1A2 or A1A3, etc. Currently, the only way we can do this is by crossing every individual plant and recording by hand.
Any information you could share on how this can be done would be greatly appreciated! I'm assuming the first step would be to map the genome of this plant?

The "easiest" way is to do a partial genome map out of several various wild types, vs the phenotype you want. You will need to do at least 10 of each to have a good idea what normal mutations/polymorphisms are vs what has as a result the phenotype you want. Problem is, you need a general idea of where in the genome your mutation is.
You could use interfering oligonucleotides to suppress the expression of your phenotype to more easily find the gene and or location you want.
