I can't raise the phage titer no matter what I do. I added 7 ml TSB, 2 ml phage lysate and 1 ml Y. ruckeri and incubated for 1 night, but the phage titer did not rise. I added 100 microliters of phage lysate to 4 ml of 1-night Y. ruckeri and the titer did not rise. What should I do to raise the phage titer?
Why don't you pick one of the plaques from a plate, you can grab it with a Pasteur pipet, and add to a growing culture (make a 1:100 dilution from an overnight culture) and let that incubate for 4-8 hours until you see lysis. Also do you know the cation requirements for your phage? Some phages need Mg or Ca to stabilize, so you might try adding 1mM CaCl2 and 10mM MgSO4 to the growing cells.
Inoculate your virus into the host culture in the exponential phase.
