Dear all, I would like to know if a particular Arabidopsis gene might be downregulated by miRNA (or siRNA or tasiRNA). How can I identify putative regulators?
Difficult question. I can suggest a strategy, although I donot know the feasibility of the same. If you know the whole gene sequence or accsn# then you might use this online tool to find putative miRNAs. After than you need to transfect the miRNA into the cells and check by qRT or WB whether the gene of interest gets down regulated or not. I think Elisa is searching the miRNA (unknown) against the known gene. If I understood the problem correctly, then it might be that she would like to evaluate or identify the miRNA.
