Now, i am going to find the ligand of molecular X. We are preparing to overexpress the molecular X in cells , and get the cell membrane to co-IP. But someone has told me the ligand and the molecular X has low affinity, and co-IP will not be able to pull them down.

But other scientists has improved the ligands of molecular X existed by the immunofluorescence methods.

So, my problem is, how can i find the low affinity ligands? Can i guess some potential ligands and overexpress them, and then to do the co-IP? Is this method will help?
