Hi!

I am looking for an efficient method to remove debris and dead cells in a single cell suspension from human tumor. After mechanical and enzymatic dissociation using GentleMACS system, I get only 10% viable cells. I tried the dead cell removal kit (MACS Miltenyi) however it was not very efficient (I got 24% viability). I am going to perform single cell RNA-seq and it is very important to have at least 70% viable cells.

Does anyone have experience in preparing tumor samples to scRNA-seq and suggest me another protocol?
