As I am working with Chlorella vulgaris stain. During fluorescence microscopy analysis the cells were showing little fluorescent intact and same time floating under the microscope that I am unable to take images.
Please find attached articles related to Nile red staining. We are using same protocol. Hope it will work for you. Though I prefer BODIPY dye instead of Nile red.
About little fluorescence, the use of solvent for dissolving nile red dye, the incubation time and temperature is important. I have attached a link hope it might help you.
