I am going to profile microRNAs by small RNA sequencing. The starting material that is going to be used for microRNA isolation is too small. So we decided to isolate small RNAs by three different kits (miRVana, PureLink, Qiagen - all for small RNA isolation), assess the RNA by Bioanalyzer, and next to pre-amplify small RNAs by two different kits (Global MicroRNA Amplification Kit from SBI and NCode MicroRNA Amplification System from invitrogen). We performed these isolations and pre-amplifications and unluckily got nothing promising to proceed to small RNA sequencing.

So please let me know of your suggestions/recommendations and comments if you have any good experience on isolating small RNA from very small amount of starting material and small RNA sequencing of the resulting small RNAs.

Thanks in advance,

Sharif
