I want to dissolve Paclitaxel(which is already dissolved in DMSO) in Cremophor EL and Dehydrated alcohol for my animal studies. I will be giving IP injections to nude mice.
You also need saline to make a solution for injection.
I always prepared it from powder, but I'm sure it will also work with DMSO stock:
The solution is 1 part ethanol, 1 part Cremophor EL, 8 parts saline.
Dissolve the PTX in ethanol. Add Cremophor EL and mix well. Add saline and vortex. The solution should remain clear after you add saline. Sometimes the solution it not stable and can precipitate after a couple of hours. I always prepare it fresh before injections. I injected IV though, I guess if you're injecting IP you can also use a cloudy suspension.

Last time when I made injections. First I mixed cremophor EL and ethanol. Into this solution I dissolved PTX(which is already dissolved in DMSO) and make up the volume with water. The suspension was little bit cloudy. I am not sure this the right way to dissolve the drug.
The PTX needs to be well dissolved in ethanol before you add cremophor, or it will precipitate. And after adding saline vortex immediately for about 20-30 seconds at least. Use saline, not water, so the solution will be isotonic.
I too had the same difficulty in disolving my Docetaxel drug which was already dissolved in DMSO. Taxnes are hydrophobic and hence dificluty in disolving in water based solvents. Before you could dissolve DTC/ paclitaxel in saline, add few microlitres (maximum of 1.5%) of polysorbate 80 (tween 80) or tween 20 which will greatly enhance the solubiliyt of taxanes in saline and can be used for IP injections in mice.
