Dear Vamshi... for the preparation of any milk product one has to do the standardization of milk for Fat and Solids-not-fat  (SNF) content and the same needs to be declared on the label ( This product is prepared from Toned milk or skim milk and so on.,) so that the product composition and other  properties would be maintained  uniformly, so the effect of composition of milk on product quality is nullified.

Coming to your question:

1. By organoleptic (sensory evaluation) one can make out the type of milk by its color.  Cow milk is yellowish creamy white in color, while  Buffalo and Goat milk  are of  creamy white in color ( Because: beta- carotene pigment  gets converted to colorless Vitamin- A, this conversion efficiency is less in cow). This color difference is  not an ideal test to differentiate in many cases (diluted or processed milk).

2. Milks of the closely related species can be differentiated by the precipitin test using antisera to blood serum proteins. with this one can make out  the "Fresh or  reconstituted milk  Reference: F. 3. Hansa test helps to detection of adulteration of cow milk with buffalo milk ( by ICAR-National Dairy Research Institute)

4. ICAR-National Bureau of Animal Genetic Resources has developed a PCR based  test; this test is able to differentiate genomic DNA of cow and buffalo origin utilizing size differentiation of PCR amplified products from single set of primers simply by agarose gel electrophoresis.

if these  samples not adulterated you can measure the fat % of each in addition to organoleptic examination

Goat milk has a higher dry matter and fat is characteristic with the presence of caproic acid, caprylic acid ...

The differentiation between  the three types of milk will be according to their fat content , color and smile the goats milk contain gouty flavor and low fat content as cows milk also have low fat however buffaloes contains high fat content

the color of the cow milk is moderate from white to light brown but, the scientist when asked about the color of the milk they ask , it is colorless.when compare with baffalow , the color is whit with high rate of lipid. the milk of goat is watery and white in color less than baffalow milk and low percentage of lipid.

To differentiate milk from cow and goat, we used in France a commercial kit based on specific antibodies detected by immuno radial diffusion (IDR) . Sensitivity and specificity are very  high and the method is cheaper

Differentiation based in PCR amplification of species-specific mitochondrial DNA (mtDNA) targets could give you an accurate response to your problem.

If you don't have a clear idea about the milk you are using from the supplier, you must check for the possibility of having an undetermined mixing of milks from cow, buffalo, goat (and maybe ewe, or any other species). The only solution would be to use either PCR or immuno test IDR as suggested by Bernard Poutrel or by  Maria Do Rosario Marques. Given the effects of female diets on milk composition, basic fat or colour or organoleptic analyses cannot be conclusive, especially if milks were mixed or adulterated

Can I suggest that you could try and source milk from specific farm suppliers to give you the species type that you want, rather than buying from a commercial source.

Laboratorical eamination determinates the rate of fat in each type of milk very important.

We have developed an ELISA test which can detect adulteration of cow and buffalo milk. We can detect 5 % mixing of cow milk in buffalo milk or 5 % mixing of buffalo milk in cow milk in about 15 minutes. This is a simple visual test and does not require any equipment. There are several testing methods available with testing strips that could give you verry rapidly a definitive answer. However a trustworthy supplier seem the best method to me.
