I have a Shimadzu LC-triple quad with a dial on the side of the ion chamber. I am using the instrument for targeted MRM analysis while using an internal standard to account for any recovery issues. I am looking for a strategy to hone in the capillary. What criteria should I be using to determine the proper positioning on the dial? Can I expect the positioning to effect peak shape, instrument response, tailing factor, absolute area counts?
This dial is to adjust the spray position and is dependent on the flow rate from the LC. Recommendations are that for flow rates between 0.2 and 0.5ml/min the spray position should be between -1 to + 2mm and for flow rates of 1ml/min +3mm or greater. The position should be set experimentally to give maximum abundance.
Peak shape or tailing factor will not be affected by the spray position.
