Hi Bhagya, I think if you want to check immuno dominant activity of an antigen, an obligation is using LBA (ligand binding assay). in LBA, the most common Binder is antibody. So you have to use Antibody but the reaction of intended antigen and the antibody must be follow with help of label, tag or probe. Among the various label, enzymes are the most common and applicable molecule, so finally ELISA will be the best choice. of course in some situation you can use WB (western blot) and in other you can use IHC.
