Is there one specific marker just to validate the conversion to dendritic cells? I have found several panels to analyze the immunophenotype of DC's, but I would like to know if there is an option to demonstrate it just with one or two markers. Thank you in advance.
A positive staining for MHCII and CD11c should be enough to demonstrate that your monocytes have turned into DCs.
To be sure to have DCs from monocytes, look at the expression of CD1a or CD 209 that are specific markers.
To follow a maturation of these mono DC, via CD154 for example, the CD208 is really the most representative marker: the switch between CD68 and DClamp is edifying
another staining for CD14 would also be good - as monocytes are CD 14 positive and DCs are CD 14 negative
Many thanks! The different subsets of DC's make everything more difficult, but I think that I will go for CD1a and DC-SIGN, as Dr Jean suggested. Again, thanks to you all.
