How can i culture mesenchymal stem cell on collagen membrane and how can confirm adherence?

i have one piece of collagen membrane (hydro-sorb F ) ( bioimplon) and i want to seed mesenchymal stem cell on it for local application in animal model , also i have granule like material and i don not know if it will adhere to the membrane or not.(my aim is to culture stem on these granule which supposed to be attached to collagen membrane)

And if stem adhere how can confirm adherence by method other than electron microscope

you can fix the membrane on a spinal needle then hang in spinner flask containing the cell suspension
