I want to conjugate Streptavidin to the surface of gold nanoparticles and gold nanorods. The surface chemistry of gold nanoparticles is citrate whereas the surface chemistry of gold nanorods is CTAB. Any suggestion? Thanks
if u want to conjugate the streptavidin. first thing u need to remember that, whether surface of Au has either if the group like -OH, -COOH, NH2, if you had modified with the above mentioned group then you can use EDC- NHS chemistry to covalently bind Streptavidin to the gold nano particle. remember pH definitely play an role in this reaction.
Hi, I used Citrate-AuNP about 12nm in diameter. First I changed the buffer for 2mM boric acid pH 8 simply by centrifugation and resuspension. I did it twice. Then I titrated the AuNP solution with different Streptavidin concentrations: 1,2,4,8,16,32 and 50 ug/ml of AuNP for 2h at 23oC in thermoshaker (800 rpm) in eppendorf low binding 1.5 ml tubes. After 2h you can make UV-VIS spectrum measurement to check SPR maximum wavelength. You should observe shift to longer wavelength. Then add equal volume of 10% NaCl and record the UV-VIS spectrum once again. In the case of successful SA absorption the AuNP should be stable and SPR maximum wavelength should be the same as without NaCl.
