Hi. I have obtained AFM Images of polysaccharides using tapping and scanning mode. I want to calculate Young Modulus or Hertz model of elasticity. How can I calculate elasticity from AFM images or which software will be helpful? I am beginner in AFM. Need suggestions and guidance. Thanks
If you've just taken a regular image with a regular tip you can't do that yet.
You need to perform indentation experiments, at best with special tips. Please check whether your AFM has the proper control options and whether the company that made your AFM sells such tips.

I used silicon tip (2 nm) on Nitride Lever while used Bruker AFM. Mode was peak force tapping mode with a spring constant of 0.4 N/M. I am not sure if can calculate it or still need to find another AFM.
Well, if your AFM has a peak force mode you should be fine. The modulus calculation sholuld then proceed as described e.g. in
