I am trying to develop anti-drug antibody (ADA) assays. Typically one uses an acid treatment to break up any existing antibody-drug complexes prior to testing for the ADA. Does anyone know of another way to break the complexes besides acid that would still be compatible with detection of ADA?
You have conjugate drug to antibody through cleave-able bond and not by covalent bond. For this you can refer G. T. Hermanson book on Bio-conjugate Techniques.
Thanks for the answers everyone. I am intrigued by Falk's idea of 3M MgCl2. Since my drug, in the ADA complex, is an antibody therapeutic I need to figure a way to remove the MgCl2 to let the antibody bind to a labeled version of the drug for detection purposes. Perhaps size exclusion chromatography or something like it?
