I am calculating the encapsulation efficiency of drug loaded polymeric nanoparticles. These nanoparticles are soluble in acetone. So, acetone is used as the solvent for measuring the UV-absorbance of our sample in order to calculate the concentration. However, during measurement, the UV spectrum shows irregular peaks even after proper baseline correction, making it difficult to accurately determine the absorbance and thus the drug concentration. Can anyone suggest how to troubleshoot this
You have to chose a liquid such that your material should not dissolve in that liquid. Your material should be suspended in that liquid uniformly. You first take absorbance of liquid only. Then take absorbance of liquid plus your dispersed sample. By this process you can easily calculate concentration as well as absorbance.
Try using a cosolvent to dissolve the sample (e.g., water + acetone or ethanol + acetone in a different ratio).
