I'm working on quantifying cell dynamics on a three-dimensional matrix and rely heavily on three-dimensional microscopic images. However, I was never able to capture high-quality images of my cells on  3D fibrous matrices. I understand the focusing error caused by the uneven planer surface, as well as the failure in Z stacking. Please provide me with a feasible protocol for capturing cell morphology encapsulated in 3D fibrous matrices?
