Na/k acetate is used to neutralize the nucleic acid following alkaline lysis where Na/K binds with the nucleic acid followed by alcohol precipitation.
Na/k exist with DNA following elution or we get purified DNA alone?
Hello,
While you doing alcohol precipitation DNA get precipitate, Remaining other Na/K ions dissolved in alcohol. Finally you will get purified DNA
I think you end up with the Na+ or K+ salt of DNA, not the free acid.
I have got two different comments from two researchers. is there anyone who help me to draw a conclusion?
I would agree with Adam. I don't believe the alcohol precipitation would remove the salt. You could use a Sep-Pak C18 column to bind and wash the DNA, then eluting with Methanol. This should remove any salt if you are concerned.
