I have Downstream gene variant as the type of variant in one of the gene of my interest affected by mutation. But now, I am having trouble interpreting how the sequences affected after the gene sequence can effect a function of gene upstream to a affected region. I know this is possible if the gene is trans regulated.

Any other possible explanation or reasoning will be very helpful.
