I have a decrease of 1/3 to 1/2 in the gene expression of gluconeogenesis enzymes (Glucose-6-phosphatase, Fructose-1,6-bisphosphatase, Phosphoenolpyruvate carboxykinase and Pyruvate carboxylase) in mice liver between my test condition and my control condition and I would like to know if it can lead to a reduction of the flux of the pathway or not? Also is one of these enzymes more critical than the others in the pathway?

To further elucidate the possible metabolic outcome associated with changes in the gluconeogenesis pathway, protein expression of these enzymes is warranted. As for the second question, Fructose-1,6-bisphosphatase (FBP1) is the rate-limiting enzyme in gluconeogenesis.
I agree with Rola, however you could make the claim that these data suggest that gluconeogenesis is impaired in the in the test subjects. Protein quantification will likely mirror these results and strengthen your argument.

What about protein level? Transcription level is not always correlated to translation level. Furthermore enzyme activity might also be regulated at the protein level. So it's difficult to conclude anything from your data.
