hello every one, in gene targeting if the flank homology arm contains several mutaion does this dismatch affect recombination efficient? the whole sequence homology ratio can reach 90% also.
Obviously, the mismatch will affect.
Depends on the organism. Neisseria and Strep don't have much problem. Anything else, probably.
Even in the Neisseriacea there are big differences and 10% sequence difference can drop the transformation frequency by several log10.
