I am in doubt about different methods of preparations and calculations. While I performed it I got turbid material after denaturation by heating at 57 C for 3 minutes. So the absorbance at 416 nm is so high. Which is to prefer 416nm or 660 nm for measure absorbance?

There are reports on the significant protein denaturation inhibitory activity with good protection against erythrocytes membrane lysis indicating anti- inflammatory potential. If the absorbance you obtained at 416 is so high you should first of all try higher wavelengths of over 600 and above and the one that gives you the best result. consult this ref. https://www.ncbi.nlh.gov
