For the histopathological investigation of the hippocampus by H&E staining, we had cut slices of 7 µm thickness manually with a microtome. As many as slices can be cut from a single hippocampal tissue and out of that best 2-3 slices can be accommodated on a glass slide, which can further be stained for further examination to obtain the correct results. The least number of sections to count the neuronal cells in each animal can be 2. However, more sections should be incorporated to confirm the results.

The magnification of 400x under a light microscope is the best for counting and examining the pyramidal neuronal cells with conical or round intact neuronal cells possessing noticeable nuclei. Further, deformed and disfigured neuronal cells, with fragmented nuclei, irregular distribution, hyperchromatic or vacuolated nuclei, and darker shade denoting neurodegeneration can also auspiciously be observed under this magnification.

The slides can be observed under 200x and 100 x as well to locate the exact region of CA1, dentate gyrus and CA3 of the hippocampus.

But as we go deep in parafinized block to get more section, loss of tissue is evident, immediately, preferably of the hippocampus, after placing the slice on slide. So better way is to cut minimum number of section (10-12) and select 2-3
