I did infection for Galleria mellonella by injection 20 microliter from different bacteria (MRSA300, NRS71,and VRSA4) and used one group as control by injection 20 microliter from PBS into haemocael of each worm and I incubated the worm at 37c0 in incubator. i observed after 20 hr all groups of the worm die even the control. any one have a clue for this observation?
Hi, did you seal injection site with liquid paraffin? Bleeding is important.
Agree with Arash, from the bleeding microbes could be spread.
Perhaps your larvae were contaminated with Proteus sp. or other bacteria even before you injected your bacteria.
Never had problems with bleeding so far, but isn't 20 microliter quite a lot to inject, probably you could try 5 or 10 microlitres.
20 microliter is far to much to inject. MRSA is very lethal pathogen so you need to adjust the concentration of the bacterial suspension and use only 5 microliter to inject.
