I want to study the physiology of the larvae of a moth, and I must cut the cross section of the body of the larvae, and see the fatty granules of the body. Dear researchers, What is the simple way to create a transverse section of the larvae?

Just a few questions:
1. Are you wanting to see the fat body cells, or the lipid droplets within the cells?
2. What type of microscopy will you employ (brightfield, fluorescence?)
3. Which stain did you plan to use?
I would maybe suggest fixing the larvae (e.g. in 4% fresh paraformaldehyde) and embedding in paraffin (or similar), then sectioning and staining. You can visualize fat cell morphology with Nile Red or Tubulin antibody (both fluorescence stains), or probably even hematoxylin & eosin (brightfield).

The simplest way to get a clean cut through the body is to deepfreese it, and make a cut with a surgical scalpel.
If you stain the cut surface with a histological dye for fat tissue, you should be able to see what you want.
