I am currently collecting conditioned medium, where once the cells have reached confluence I will replace the culture medium with new complete medium and incubate 24h before collecting. But I have seen other protocol use serum free media instead of complete media.
Using low serum (0.2%) or serum-free medium for the conditioning step makes analysis of secreted components easier and more accurate.

Cells are usually seeded with the complete media and allowed to attach and recover for certain period of time. Since serum contain a multitude of factors which may cause changes within the treated cells, serum free media should be used for the treatment step. This also ensures that the conditioned media is free of any agents coming from the serum that can hinder the downstream processes

Serum free medium can be used to synchronise cells: serum starvation induces cell cycle synchronisation which is a prerequisite for some experiments
Nur Mohd Faizal Yes ! that is a good thought but with serum media you can not identify the single causal factor for your results (and chain effects). As such the cells outside the body are not 100% in 'home' environment. All the cell culture experiments have this one disadvantage and hence animal studies are thought to be the higher level of research studies.
By use of control and treatment groups, we are able to control the entire experiment and infer the results better !
