I have the lungs, spleen and lungs of=f the BALB/c mice with me and I want to proceed for H&E staining of these tissues. But I would like to know whether any specific section from these tissues is to be used or any random small section from these organs will do.
Dear colleage, in my experience as a veterinary pathologist, M bovis does not have predilection for any area of spleen, lungs and liver, althoug when you select the sectios of tissue for fixation you should include the border zone between the aparently normal and the lessioned tissue, there are the most important changes with diagnostic value, I guess that most be similar with M tuberculosis, by the way, the best animal for isolation of M bovis from biological samples is guinea pig
I think the important thing is that to have the well-stained section for imaging rather than specific area, anyhow the best section can be provided by middle part of para fin block.
I think,by placing good tissue direction in paraffin block, you get better results. the rest of staining process is similar to conventional H&E staining.
