There are many kits or reagents for serum concentration of triglyceride, however, I'm not sure those products can be used for liver triglyceride measurement.

Following is lipid extraction protocol:

1. Extract whole lipid from liver by using cholorform:MeOH(2:1).

2. After homogenizing liver in PBS, add chloroform:MeOH reagent to homogenate in 15 ml cornical tube and centrifuged at 1500 rpm for 10 min.

3. Collect lower part and repeat this process with upper layer.

4. Merge two lower parts and move to glass tube.

5. Dry for 1 day and resolve with ethanol.

I used the kit that has the same composition with Product Nos. TR22203 and TR22215 (Thermo).

The 10 ul of sample was incubated with 1 ml of reagent for 5 min following measurement at 505 nm.

I found that the absorbance of the same sample changed extremely with every measurement.

Therefore, I have doubts about application of TG reagent for serum diagnosis or experimental protocols.
