I ran a pilot study in mice using CNO in the drinking water. You need to monitor their fluid intake for several days and use their average water consumption to determine the concentration of CNO stock solution to add in order to get your desired final dose. I don't know the rat numbers of mg/kg are, but you should be able to find them.
The biggest problem I had is CNO solubility. It must be dissolved in DMSO, unless you order a water soluble version (HelloBio). Only make up as much as you are going to use. If you try to make a stock solution, it will likely come out of solution in a few days, particularly if you put it in a fridge. The other thing is that you have to make sure control animals are also receiving CNO in the drinking water. Back metabolism to clozapine is a real issue over extended periods. You need to be certain your effects are due to DREADD activation and not clozapine.
