I want to do pharmacokinetics study of tacrolimus in rats. I want to use ELISA kit for the determination of the drug concentration.
So, i would be thankful if anyone can provide the protocol in detail to prepare sample for analysis.

Dear Mr. Pathak.
As I experienced for sample preparation, the manufacturer's protocol will explain it.
Generally :
1. Collect the blood in heparized tube.
2. Then you should decide whether you want the serum or the plasma. Using cenrifugation, separate the plasma or serum, I used 6000 rcf for 4 minutes.
3. Take the serum or the plasma then process as the kit's instruction.
4. If you need to store the sample, keep in -20 degree for short term storage or -80 degree for long time storage.
