I am trying to purify a protein complex from Hela Cells Nuclear Extract
First, Hela-S3 cell was grown in DMEM supplemented with 10% FBS in dishes. The cells from an 80% confluent 10 cm dish are detached and centrifuged. The cell was resuspended in 50 ml supplemented Joklik's modification MEM(sigma)+10%FBS+1% NEAA+2mM glutamine+penicillin-streptomycin and grown in a 250 ml flask. but when the cells are in suspension culture, it forms so many clumps that I can see these clumps even with my naked eyes. I don't know why and what should I do to make the cell in better condition.
