I have obsevered several times that HEK 293 cells seem to be detached easily in comparison with other cells. Particularly, the time of trypsin response is extremely rapid (30 seconds).
I would ask that in your case HEK293 , does it show the same characteristic

Yes, it does show the same trend. HEK293 has a tendency to come off the substratum without much efforts. Sometimes, just pipetting a few times will get your cells in suspension. You may not even have to use trypsin in certain cases.

Yes, this is completely normal for HEK293 cells. Sometimes even as little as shaking the plate is sufficient to dislodge a significant portion of your cells in a dish. Best to handle them with an extra level of care!

The phenomenon you're observing is perfectly alright and it happens a lot. HEK293T cells have this ability of getting detached from the culture dish surface very easily. Especially when the confluency reaches around 60-70%. I have experienced the same thing on several occasions. Whenever you do pipetting, try to be very gentle with 293T cells and prefer not to grow them beyond 70% confluency. Or else you may try culturing them in poly-L-lysine coated 96 well dishes or culture flasks. This will improve their adherence abilities to the culture dish.

Yes it detaches very easily. So you have to be very gentle when handling them.
