We need to use this genotoxic test in Chlamydomonas reinhardtii, for that we want to know which is the best method to eliminate the cellular wall of the cells and more information about the analysis using flow cytometry.
Micronucleus tests (MNT) are best used in animal cells and animal based MNT are widespread world over. Using MNT in plant cells to understand the geno-toxicology is poorly known.
I have used MNT in fishes to assess geno-toxic effects of various chemicals.
Micronucleus (MN) assay is very easy and rapid test for the assessment for genotoxicity of any compound in any models (Cell lines, blood, animals, etc.,). The MN assay needs to be standardized against any models. For flow cytometry analysis, use the following references to under stand the basics.
