I'm using this kit quite often and instructions mention that frozen samples can be used. However, I think it will only work well when the snap frozen cell lysates were originally obtained with the actin stabilization buffer from the kit (containing ATP and actin stabilizers). If your samples were extracted, for instance, with RIPA and the snap frozen then actin structures won't likely survive the temperature shift required to follow the protocol.

Also spend some time trying this assay using the method detailed in this paper,

mainly because you can use table top centrifuge @15000rpm for pelleting F-actin, but on running a gel did not see any F-actin band so it's cyto-skeleton kit only in future.
