I am searching for papers about mutation to produce kinase active variant of Tec protein. I can see a lot of papers with kinase dead Tec, but I haven't been able to find one with Kinase active.
Recombinant TEC may be procured from e.g. ThermoFisher Scientific. Have a look at their homepage. As well established for receptor tyrosine kinases its an intracellular fragment and it exhibits in-vitro kinase activity. Apart from the fact that it is a fragment, the sequence is wt
As an alternative, is there a known upstream kinase with an activating mutation? One possibility if there is a suitable candidate would be to engineer a fusion protein between the upstream protein and the TEC kinase fragment with a flexi-linker, then express this as a recombinant protein.
Hello Allan Mah, I am using the kinase active Tec from signal chem, but it would be wonderful if it is possible to know the kinase activating mutation in Tec so that i could use it in in vivo assays as well.
