We are trying to create in vitro models of Foreign Body Giant Cells with the THP-1 cell line. Literature seems to be very muddy, and most in vitro models are done with primary cells. Has anyone induced the formation of multinucleated giant cells with >20 nuclei and if so, what was the procedure?
Separate question, has anyone performed re-plating of THP-1 cells before? What would be the ideal method of detachment and then would these cells re-adhere if I added them to a new flask?
Regarding your second question, to the best of my knowledge, THP-1 cells grow in suspension. Please consult the datasheets from ATCC or DSMZ.
