We are currently breeding J20 males and rTg4510 females from the Jax Lab and are having breeding difficulties. We pair for 2 weeks (we also give Lovemash throughout the breeding period). Has anyone experienced difficulties with breeding using these strains in the past and how have you remedied them?
Hi Stephen, we have never had any problems with breeding J20 mice (never tried rTg4510). What is the problem? Is it just they wont breed? Or that you are not getting the correct genotypes? If it is a genotype issue it is quite possibly emrbyonic lethal with the two genes. If they wont breed, what is the age you are setting them up at? The J20 mice will often stop breeding at older ages- or die- so we will set them up from 6 weeks of age. Also are you maintaining them as heterzygous mice?
In our lab we have experienced a few cases of male J20 mice being infertile. We are repairing the tau females (in a 1:2 fashion rather than a 1:3) this next week with the male and will see if we see any improvements.
I have called Jax and nothing between these 2 strains gave any indication of an embryo lethality.
We set up the male at about 8 weeks and the females 6weeks (as recommended by the Jax lab). The J20 is heterozygous for hAPP and the tau mice are heterozygous for CaMK2a and P301L tau

Could you please describe the breeding procedure you use for J20 mice? We maintain them as Hemizygous mice in our laboratory.
