Hi, I'm interested to know the effect on myelin sheath of myelinated nerve fibers after fixation in 4% PFA? is there any difference if we fix in 4% GA (glutaraldehyde). Thanks for your response. It would be helpful for me.
Glutaraldehyde fixation and dehydration for TEM Will reduce the spacing of the myelin layers slightly, but glutaraldehyde is absolutely necessary for good EM level structure.
dear khairunnisa, i've used PFA for fixing the spinal cord, in response to your question i should say that GA is a better fixative for EM purpose because of the preservation of tissue elements,but small piecec of tissue is necessary. and formalin is the most routine fixative in histology labs. eventually, i suggest to use PFA as a fixstive. it is very good for IHC techniques.
