I would like to work with this cancer cell line, but I found odd culturing requirements, such as working in a CO2-free environment, whereas in different publication they have been cultured in standard conditions.
I often work with SW48 cell line and there is no need for special handlings with this culture. We culture them in 5% CO2, 37´C in DMEM supplemented with 10% FBS, penicilin-streptomycin, glutamine, and sodium pyruvate . Also, no trypsin is required, they are OK if a cell scraper is used.
The CO2-free environment would be necessary if you grow them in Leibovitz's L-15 Medium. The L-15 medium formulation was devised for use in a free gas exchange with atmospheric air. A CO2 and air mixture is detrimental to cells when using this medium for cultivation.
