Recently, I am trying to isolate islets from STZ induced diabetic mice. I have tried to tweak the type of the collagenase, collagenase concentration, incubation time, stoping method..etc.. but I rarely found islets from the STZ induced mice (usually around 50 per mice at most). It looks like they are all digested.
If you guys have tried islet isolation in STZ induced diabetic mice, what could be the best way?
It depends what is the motive of isolation, if you only want to check the cell morphology than i will suggest that you can go for histology of whole tissue and use histo- morphological stains for islets. That will help you to identify the cells and also provide you the detailed information about the shape and changes of your treatment on islets
